چکیده

BIOCHEMISTRY Correction for “Confinement of caspase-12 proteolytic activity to autoprocessing,” by Sophie Roy, Jeffrey R. Sharom, Caroline Houde, Thomas P. Loisel, John P. Vaillancourt, Wei Shao, Maya Saleh, and Donald W. Nicholson, which appeared in issue 11, March 18, 2008, of Proc Natl Acad Sci USA (105:4133–4138; first published March 10, 2008; 10.1073/pnas.0706658105). The authors note that Fig. 6 appeared incorrectly. There was an error in the alignment of the molecular mass markers, and minor adjustments have been made to the assignment of caspase12 bands. The authors also note that the source of the anticaspase-12 antibody for Fig. 6 was Sigma (clone 14F7). This error does not affect the conclusions of the article. The corrected figure and its corresponding legend appear below.

Mechanism of Dronc activation in Drosophila cells.

Proteolytic processing is required for the activation of most caspases. However, recent reports have suggested that the activation of the mammalian initiator caspase caspase-9 occurs during dimerization rather than after processing. Previously, we reported that, in normal living Drosophila S2 cells, the initiator caspase Dronc is continuously processed to a 40 kDa form we called Pr1 and that, d...

The Apaf-1*procaspase-9 apoptosome complex functions as a proteolytic-based molecular timer.

During stress-induced apoptosis, the initiator caspase-9 is activated by the Apaf-1 apoptosome and must remain bound to retain significant catalytic activity. Nevertheless, in apoptotic cells the vast majority of processed caspase-9 is paradoxically observed outside the complex. We show herein that apoptosome-mediated cleavage of procaspase-9 occurs exclusively through a CARD-displacement mecha...

Autoprocessing: an essential step for the activation of HIV-1 protease.

Human immunodeficiency virus type 1 (HIV-1) expresses its structural and functional proteins within Gag-Pol precursor polyproteins. Specific proteolytic processing of the precursors by the viral protease is critical for the maturation and infectivity of viral particles. To observe the influence of autoprocessing on the activation of recombinant HIV-1 protease, we constructed different HIV-1 pro...

Hetero- and autoprocessing of the extracellular metalloprotease (Mpr) in Bacillus subtilis.

Most proteases are synthesized as inactive precursors which are processed by proteolytic cleavage into a mature active form, allowing regulation of their proteolytic activity. The activation of the glutamic-acid-specific extracellular metalloprotease (Mpr) of Bacillus subtilis has been examined. Analysis of Mpr processing in defined protease-deficient mutants by activity assay and Western blott...
